C) No booklet will be marked without the MCQ question paper
Section A 25 marks
Read the following literature carefully
1. Explain why Eosin-Methylene Blue is both a selective and differential media. 3 marks
2. Zebedee and Larisa decided to clean the surrounding of the lab. Accidentally Larisa pricked her leg with a nail.
5 days later she was suffering from high fever and the leg was swollen. When she went to the hospital,
Onyinyechi said she was suffering from Tetanus. Blandine decided to confirm the staining reaction with a
culture test. However, since the lab technicians were females, Munang and Zebedee were not too convinced
of their result and decided to use the Koch’s postulates to confirm the diagnosis. Assuming that all aseptic
techniques were respected; answer the following questions
a. Outline the steps of the straining procedure being carried out. 4 marks
b. Explain the steps Blandine will use to confirm that she is suffering from tetanus 2.5 marks
c. Explain briefly how Blandine prepared the culture media. 2.5 marks
3. A new bacterium was discovered during the MCBS 3101 practical. The bacterium was found to divide every after
30 minutes and does not exhibit a lag phase. At 7.45 am the next day, Faith transferred some cells from the
exponentially growing culture to a fresh source of the same medum of cells (per milliliter) that will be in the medium
at 9:15 am. 5 marks
4. in a tabular form, match the following tests (A) used in identifying bacteria with the following reagents, material or
enzymes (B). 6 marks
A: a) Catalase Test b) Citrate Utilization tests c) DNA Microarray d) Indole Test e) Motility test f) Oxidase test
g) Polymerase chain reaction h) Sugar (Carbohydrate) fermentation test i) Voges-Proskauer Test j) Western
Blotting
B) 1) 2,3-butanediol 2) Artificial final electron acceptor 3) Brom-thymol blue dye 4) Hydrogen peroxide
5) Durham tube 6) Kovac’s reagent 7) Oligonucleotide 8) Seperation using sds-page 9) Taq polymerase
10) Tetrazolium motility agars
SECTION B

ii) All prokaryotes are unicellular and all eukaryotes are multicellular
iii) Spontaneous mutations are induced by agents such as 5-bromouracil
iv) Bergey’s Manual of Systematic Bacteriology is a reference for viral taxonomy

vi) A bacterial infection has the same outcome in all susceptible hosts
vii) IgE binds to most cells and participates in allergic reactions
viii) Pasteurization can be used to achieve sterility
ix) Tyndallization can be used to sterilize plain water
x) Filtration is a chemical method used to sterilize sensitive pharmaceuticals
2. Write concise notes on the following (8 + 8= 16 marks)
a) Attributes of a microbe that enhances its virulence
b) Griffith’s transformation experiments
3. i) List and explain the genetic materials of bacteria (5 marks)
ii) Draw and label an IgE molecule (4 marks)
